Acrylic thickness

I’m making a box frame 18” x 10” with a deep glass spacer. The customer has asked for acrylic which I don’t do a lot of, would I need 3mm at this size to stop sag or would 2mm be sufficient?

No! 2mm flexes like crazy. I don't use 2mm acrylic on anything. I use a bit of 4mm acrylic which I source locally and that's much better and quite rigid.
